Abstract
To investigate experimentally the sGAG mixture antiproliferative effect on sclera in NPDS model.
Material and methods
In our investigation we used a mixture of chondroitin sulfates and keratan sulfate. The original substance was extracted from the transparent corneal stroma of agricultural animals. Effective concentrations ranged from 0.1% to 5.0%. Subsequent investigation of these sGAG concentrations were carried out in vivo on the model of non penetrating deep sclerectomy (NPDS) in 36 rabbit eyes.
Results
Significant nontoxic sGAG mixture antiproliferative effect on the scleral tissue was noted with 1% sGAG mixture application to the operation zone, which was confirmed by histologic investigation. High concentrations of sGAG mixture (5%) were able to induce a hyperosmotic stress with interstitial edema and a gradual biochemical and morphological changes of cells.
Conclusions
The original sGAGs mixture application to the operation zone in concentration of 1% is safe, it decrease the scarring of filtration zone and intraocular fluid outflow pathways.
